two titanium frames of the chamber and close the operation field with a sterile
cover slip to avoid drying, infection, or mechanical damage of the inner layer,
i.e. the cutaneous muscle, of the unprotected side of the opposite skin. For
tissue implantation and/or other local treatments the chamber can easily be
opened again by removing the cover slip and reclosed with a new sterile cover
slip, respectively. The cutaneous muscle serves as site for implantation.
2. Materials
Except for commonly used devices, all materials necessary for the prepara-
tion of the dorsal skinfold chamber in mice are listed below with a detailed
manufacturer’s record. Evidently, all materials given are only suggestions and
may be modified for personal preferences.
2.1. Dorsal Skinfold Chamber Preparation
2.1.1. Facilities and Apparatus
1. Laminar flow hood (Faster S.r.I., Ferrara, Italy).
2. Dry Sterilizer (Model IS-350, Inotech, Intergra Biosciences, CH-8304
Wallisellen, Switzerland).
3. Dissecting microscope (Wild, 6-50×, type 334790, CH-9435 Heerbrugg, Switzerland).
4. 2 Flat custom-made thermal pads (WISAG, CH-8057 Zurich, Switzerland).
5. Halogen lamp with two flexible swan-neck light transmission tubes (Intralux,
150H, Volpi AG, CH-8902 Urdorf-Zürich, Switzerland).
6. Custom-made skin spreading device. This consists of a heavy metal base and two
flexible swan-neck tubes for applying tension to spread out the mouse skinfold,
prior to the fixation of the back titanium frame of the dorsal skinfold chamber to
the skinfold of the mouse (Workshop, Department of Clinical Research, Univer-
sity of Berne, CH-3010 Berne, Switzerland).
2.1.2. Drugs
1. Isotonic sodium chloride (0.9% NaCl solution injectable).
2. Anesthesia: Mixture consisting of isotonic sodium chloride, ketamine hydrochlo-
