Apparently from drawing, at amplification of fragments of DNA, on matrix
Вас.аnthracis pieces Ч-7, 81, 71/12 were synthesized 3 specific fragments that testifies to their
accessory to a sort bacillus and to a kind having in genome two plasmids. At the same time at
acapsular strains bacillus the Siberian ulcer (55, 34F
2
) synthesis of 2 fragments was initiated,
and with genomes heterological kinds and sorts primers did not communicate and did not
initiate synthesis of fragments of DNA. On a matrix again allocated isolates, and also at
museum strains were synthesized on 3 amplycone various molecular weight which coincided
by this criterion with amplycones known pathogenic strains.
From results of these experiences it is visible, that primers revealed loci of DNA of
microorganisms belonging to a sort bacillus, and presence plasmids рХО1 and рХО2 testified to
pathogenic strains the activator Siberian ulcer infections, that is allowed to differentiate
pathogenic and vaccines strains, and also isolate.
Bottom amplicone is genus specification that is this locus contains at all bacillus. For
example Bacillus cereus. But at them are absent plasmids and consequently two top amplicons
at them are absent. Pathogenic strains Bac.anthracis both, on the contrary, have plasmids рХ01
and рХ02 and on it on their matrix two are synthesized more amplicons which on molecular
weight it is more than patrimonial amplicons.
In our researches three have been investigated isolate, allocated in it, 2004. Isolate
«Зел» has been allocated from beef in a meat-packing plant of a city of a Zelenodolsk of
Republic Tatarstan. From meat-packing plant territory samples also soils have been taken. PCR
– indication of these tests has shown, that isolate allocated from meat contains pathogenic
strains Bac.anthracis. In tests of soil the activator of the Siberian ulcer has not been found out
(a track 11).
Same year, in one of economy of the Ulyanovsk area, two have been allocated more
isolate: from a corpse of a pig and soil on a farm where the pig has fallen. Research of these
tests with the help multiprimers PCR has shown, that in them contain pathogenic strains
Bac.anthracis, with a full set plasmids (tracks 13 and 14). For check of reliability PCR have
been in parallel investigated known pathogenic isolates Bac.anthracis, allocated for territories of
Tatarstan and Chuvashiya in 1972 and 1971, accordingly.
Further, for finding-out of an origin of these strain-isolates we had been carried out
researches on genotyping, with the help sequences amplicons from 8 marker loci: A, В1, В2,
С1, С2, CG3, рХ01 and рХ02. Are more low resulted deciphered (sequencing) nucleotides
sequences of these olygonucleotids.
By us have been investigated 6 isolate the activator of the Siberian ulcer:
«Зел» - it is allocated from beef in a meat-packing plant of a city of a Zelenodolsk; №22
– isolates museum, it is allocated from a corpse of a sheep in Republic Tatarstan in 1972; №36
– isolates museum, it is allocated from a corpse of a sheep in Republic Chuvashiya in 1971; У1
– it is allocated from a corpse of the fallen pig in the Ulyanovsk area in 2004; У2 – it is
allocated from farm soil where above specified pig has fallen; 55 – Bac.anrhacis strain 55,
vaccinal, acapsular, has no plasmid рХ02. Chromosomal DNA of these isolates and strains have
been investigated on eight marker to loci. For this purpose on DNA matrix isolates and strains,
by means of steam rooms, complementary to data marker to sites primers, were synthesized
olygonucleotids (amplicons). Further these amplicons were exposed segmentation on automatic
seguenater. By means of the special computer program seguens were analyzed on structural
structure. The given analysis has given the following picture of genetic relationship studied
strains and isolates: strains B. anthracis.
